고압력 X선 물리연구실

High Pressure X-ray Science Laboratory

우리 실험실에서는 고압과 X-선 기술을 사용하여 생물 및 연성 물질 시스템을 연구합니다. 고압력 극저온 냉각 기법으로 불리는 기술은 생물물리와 X-선 분야에서 고분자 결정의 손상을 방지하기 위해 개발되었습니다. 이 방법은 Ligand-단백질의 상호작용을 안정화하고, 단백질 구조 내부의 압력에 의한 효과를 연구하며, 효소 반응의 중간과정을 포착하는데 성공적으로 적용되었습니다. 우리는 앞으로 고압 냉각 기술을 사용하여 유리질 상태의 물의 상 변화와, 물과 단백질 사이의 상호작용, 그리고 극저온에서의 단백질 구조 및 기능을 연구할 계획입니다.
Our lab studies biological and soft matter systems using high-pressure and X-ray techniques. A high-pressure cryocooling method was developed for crystal cryoprotection in macromolecular crystallography. In addition, the method was successfully applied to stabilize ligand-protein interactions, to study the pressure effect on the structure, and to entrap intermediate enzymatic states. Using the high-pressure method, we will study the phase behaviors of glassy water, water-protein interactions, and protein functions and dynamics at cryogenic temperatures.
